Disturbances of glycine or D-serine metabolism leads to severe neurological deficits ranging from hyperekplexia to neurodegeneration. In a recently initiated collaborative study, the high-affinity D-serine transporter Asc-1 (SLC7a10) has been identified as novel and major regulator of glycinergic transmission and glycine metabolism in vivo. Although deletion of Asc-1 leads to a fatal hyperekplexia-like phenotype in mice, it is still unclear how Asc-1 and other neuronal and glial transporter cooperate in regulating glycine and D-serine homeostasis.
